Active induction of EAE in mice model tutorial

Reagent Preparation:

1. Dilute MOG peptide in PBS (10mg/ml; stock):
   • MOG35-55 peptide is diluted in PBS to make a stock solution at a concentration of 10mg/ml.
   • 200ug of this solution (20ul) will be used for each mouse.
   • Store at -70°C.
2. Mix M. tuberculosis with CFA:
   • Mix 100mg of M. tuberculosis with 10ml of Complete Freund's Adjuvant (CFA) to get a concentration of 10mg/ml.
   • Store at 4°C.
3. Dilute Pertussis toxin in PBS:
   • Dilute 50ug of Pertussis toxin (PT) in 50ml of PBS to get a concentration of 1ug/ml.
   • 200ng of this solution (200ul) will be used later.
   • Store at 4°C.

Emulsion Preparation:

1. Draw CFA containing M. tuberculosis:
   • For example, for 10 animals, 500ul of this mixture is drawn into a syringe.
2. Prepare MOG peptide per mouse:
   • For example, for 10 animals, 2mg (200ul of the 10mg/ml stock) is used.
3. Adjust the MOG peptide volume with PBS:
   • Add PBS to the MOG peptide to equalize the volumes of MOG and CFA (1:1 ratio). For 10 animals, 200ul MOG + 300ul PBS.
4. Mix MOG and CFA using syringe connectors:
   • Connect the syringes containing MOG and CFA, and mix them thoroughly without introducing air bubbles.
5. Check the emulsion:
   • Proper emulsification is indicated if the contents do not separate when the plunger of one syringe is slightly pulled back. Mixing should be done on ice to prevent protein denaturation and it takes about 4-5 hours.
6. Transfer and prepare for injection:
   • Once emulsified, transfer the mixture to one syringe, disconnect the connector, and attach a needle for injection.

Immunization:

1. Inject MOG/CFA Emulsion:
   • Inject 25ul of the MOG/CFA emulsion into each mouse, on both sides of the mouse's shoulder and the upper part of the hip (50ul per mouse in total). These sites are chosen because they are close to the lymph nodes, where immune cells will encounter the antigen.
2. Inject Pertussis Toxin:
   • After injecting the MOG peptide emulsion, inject 200ul of the prepared Pertussis toxin intraperitoneally (i.p.) into each mouse.
3. Second Pertussis Toxin Injection:
   • Two days later, administer a second i.p. injection of 200ul of Pertussis toxin to each mouse.
4. Monitor Disease Progression:
   • After immunization, monitor the mice daily for clinical signs of EAE. This is usually done using a scoring system where 0 is healthy and increasing numbers represent increasing severity of disease symptoms.

시약 준비

1. MOG peptide를 PBS에 희석 (10mg/ml; stock) -70℃ 보관

  200ug/20ul

2. CFA 10ml에 M.tuberculosis 100mg을 혼합 (10mg/ml) 4℃ 보관

3. Pertussis toxin 50ug을 50ml PBS에 희석 (1ug/ml) 4℃ 보관

200ng/200ul

 

Emulsion

*MOG peptide와 CFA의 volume은 1:1로 섞어 mouse당 100ul씩 injection.

1. M.tuberculosis가 포함된 CFA를 주사기로 필요한 양만큼 뽑는다.

  Ex)10마리일 경우 500ul를 뽑는다

.

2. mouse당 MOG peptide를 200ug씩 준비한다.

Ex)10마리일 경우 2mg이므로 200ul를 사용

 

3. MOG와 CFA volume이 1:1로 섞기 때문에 같은 양으로 맞추기 위해 MOG peptide volume을 PBS를 사용하여 맞춘다.

Ex)10마리일 경우 200ul MOG + 300ul PBS

 

4. volume을 맞춘 MOG를 새로운 주사기로 뽑는다. MOG가 들어간 주사기와 CFA가 들어간 주사기 사이에 syringe connector를 연결한다. 이때 공기가 중간에 들어가지 않도록 주의한다.

1ml 이상일 경우 3ml 주사기 사용.

 

5. 연결된 주사기를 한번씩 밀어가면서 섞는다. 단백질 변성을 막기 위해 얼음을 중간에 뭉쳐서 섞는다. 대략 4-5시간정도 걸림

Emulsion 정도의 확인은 한쪽 주사기 손잡이를 살짝 당겼을 때 안쪽에 내용물이 올라오지 않고 그 상태로 유지되면 emulsion이 잘 이루어졌음

 

6. 충분히 emulsion이 되었다고 판단되면 안쪽의 내용물을 한쪽 주사기로 다 옮기고 connector를 분리한 후 내용물이 들어있는 주사기에 needle을 연결한다.

3ml 주사기로 emulsion 했을 때에는 connector 분리 후, 1ml 주사기를 다시 연결하여 나눈다.

 

7. needle을 연결한 후 공기를 제거하기 위해 주사기를 당긴 후 아래쪽으로 내용물이 내려오도록 친다. 공기가 안쪽에 있을 경우 공기압력으로 인해 injection시 주사기에서 emulsion한 MOG가 계속 흘러나온다.

 

8. 공기를 다 뺐으면 마지막으로 공기 압력과 emulsion 상태를 체크하기 위해 100mm dish에 물을 채운 후 주사기를 조금 밀어 물에 떨어뜨려본다. 이때 내용물이 계속 흘러나오면 아직 공기가 안쪽에 있다는 것이다. 그리고 물에 떨어진 emulsion된 MOG가 퍼지지 않고 방울 상태로 유지된다면 emulsion이 완벽하게 된 상태이다.

 

Immunization

1. emulsion이 다되어진 MOG를 쥐의 어깨 양쪽과 엉덩이 위쪽에 각각 25ul씩 injection한다.

빗금 쳐진 부위는 lymph node가 있는 위치.

2. MOG peptide injection 후 pertussis toxin을 200ul씩 i.p. injection한다.

3. 2일뒤 2차로 pertussis toxin을 다시 200ul i.p. injection한다.

4. 매일 clinical score 측정
